Plants now showed a drastic reduction in growth, likely attributable to an uncontrolled overaccumulation of metals, and the vast majority died prematurely ( Fig. The consequences of this mislocalization of IRT1 were dramatic for the plants. When we looked at the localization of the corresponding mutated IRT1 protein in root hair cells, we found it readily at the plasma membrane. To evaluate if the localization of IRT1 in early endosomes was a consequence of monoubiquitin-dependent trafficking, we created a mutant version of IRT1 in which the amino acid residues that are posttranslationally modified with ubiquitin are modified. Monoubiquitination of plasma membrane proteins was indeed shown to trigger their internalization and their sorting into later endosomal compartments on their way to the vacuole/lysosome. The role of monoubiquitination in the context of plasma membrane proteins was reported years ago in several seminal articles from other model organisms ( 2, 3), guiding us in our quest for the role of IRT1 ubiquitination in plants. We confirmed that IRT1 was indeed ubiquitinated in plants and, using a set of specific antibodies that recognize different forms of ubiquitination, we observed that IRT1 was, in fact, decorated with several monoubiquitin moieties in vivo. This notably requires the covalent linkage of ubiquitin chains under the K48 form to the target protein. Ubiquitination is a posttranslational modification that has multiple roles, but it is mostly famous for tagging soluble proteins for degradation by a multiprotein complex called the proteasome. When searching for factors controlling such a dynamic behavior for IRT1, we came across a usual suspect in cell biology: ubiquitination. More generally, our work demonstrates the existence of monoubiquitin-dependent trafficking to lytic vacuoles in plants and points to proteasome-independent turnover of plasma membrane proteins. Together, these data suggest a model in which monoubiquitin-dependent internalization/sorting and turnover keep the plasma membrane pool of IRT1 low to ensure proper iron uptake and to prevent metal toxicity. We also prove that IRT1 is monoubiquitinated on several cytosol-exposed residues in vivo and that mutation of two putative monoubiquitination target residues in IRT1 triggers stabilization at the plasma membrane and leads to extreme lethality. Using pharmacological approaches, we show that IRT1 cycles to the plasma membrane to perform iron and metal uptake at the cell surface and is sent to the vacuole for proper turnover. IRT1 is unexpectedly found in trans-Golgi network/early endosomes of root hair cells, and its levels and localization are unaffected by iron nutrition. Here, we demonstrate that overexpression of Arabidopsis thaliana IRT1 leads to constitutive IRT1 protein accumulation, metal overload, and oxidative stress. So we know nearly all adult female Razorwhips live and nest at Wingmaiden Island (with the exception of Windshear), but we also know no male lives there to prevent them from performing filial infanticide. Shredstone Walls and Thorstopia are common meeting places. Theory 6: Razorwhip males live in groups females travel to find them. Now sea slugs can refer to a number of different types of animals, but normally, and particularly in Norway, sea slugs refer to nudibranches, or shell-lacking sea snails, which the Razorwhip can only get by dipping into the ocean, so I assume they are quite apt at swimming. In fact, it's their only weakness, which leads me to believe that it's really the only thing they eat and they can suffer it taken away from their food source. Razorwhips are said to have a diet of sea slugs. Theory 5: Razorwhips are excellent swimmers. They can cling to things with their tails very well, but they might also use their wing claws to aid in this as well. They are forest dragons that appear to nest in trees, but they only have two claws on each foot to cling to these trees. Razorwhips have dexterous wing claws that must be used for something, and clinging to trees makes the most sense to me. Theory 4: The Razorwhip's wing claws are used for clinging to trees. Maybe the scale grows in as the Razorwhip develops its eyesight. But the blind Razorwhip hatchlings don't have this. We see in the show that Razorwhips are diurnal, so this might be important to protecting their very large eye. Adult Razorwhips have a scale above their eyes that some fans have disliked for having the appearance of "eyelashes." I think this scale might have a use, though I think it's used to shade the eye in bright light. Theory 3: The scale above the Razorwhip's eye shades it from the sun, which grows in once they gain their sight. In order to put up with such a neighbor, I have to believe Razorwhips don't have the strongest noses. Also, in Dragons: Rise of Berk, you can find Razorwhips at Windy Pass, a place known for being well-populated with Gobsuckers, very stinky dragons that are always producing methan. For one thing, they don't have split tongues like most other dragons, which are used to enhance a reptile's sense of smell. And they need this vision because I don't think they have the best sense of smell for a dragon. This makes me think that Razorwhips have pretty strong vision. The Razorwhip has rather big eyes, and some even have purple eyes, which as stated before when discussing Woolly Howls, is an eye color useful for seeing in the dark. Theory 2: Razorwhips have great eyesight but not a strong sense of smell for a dragon. Whenever Windshear looks at something behind her, she ends up putting her head down to look between her legs, so I have to believe that's a blind spot. Not only do I think this because the Razorwhip seems to have a good vision range, but there is actually evidence for this. Theory 1: The Razorwhip's blind spot is behind it.
